Tumor Necrosis Factor alpha (TNFa)

Tumor Necrosis Factor-alpha (TNF-alpha) is a pleiotrophic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF superfamily of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-alpha also plays a role in the induction of septic shock, autoimmune diseases, rheumatoid arthritis, inflammation and diabetes.\n\nApplications: \nSuitable for use in ELISA, Immunohistochemistry, Western Blot and Neutralization. Other applications not tested.\n\nRecommended Dilution:\nImmunohistochemistry: Cryostat sections. Recommended to dialyze out the trehalose prior to staining.\nOptimal dilutions to be determined by the researcher.\n\nActivity: \n(same/more than)1x10e5 neutralizing units/mg protein. Neutralizing capacity has been determined by a cytotoxicity assay in which one unit of TNF-a is assumed to correspond to 50pg of TNF-a protein.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute by adding sterile 40-50% glycerol, ddH2O. Aliquot and store at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
