Tumor Necrosis Factor alpha (TNFa)

Tumor Necrosis Factor-alpha (TNF-alpha) is a pleiotrophic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF superfamily of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-alpha also plays a role in the induction of septic shock, autoimmune diseases, rheumatoid arthritis, inflammation and diabetes.\n\nApplications:\nSuitable for use in ELISA, Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nImmunohistology (frozen and paraffin): 1:50. Non-specific background staining is seen in connective tissue. \nFlow Cytometry: 1:50\nWestern Blot: 1:50\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes human native and recombinant TNF-alpha. Reacts with soluble (17kD) and membrane (26kD) human TNF-alpha. Does not crossreact with TNF-beta, lymphotoxin or receptor bound TNF-alpha. Species Crossreactivity: mouse, guinea pig and rhesus monkey.
