Tumor necrosis factor is a multifunctional proinflammatory cytokine, with effects on lipid metabolism, coagulation, insulin resistance, and endothelial function. Tumor necrosis factor-alpha (TNF-alpha), the product of activated monocytes, is secreted by macrophages in response to inflammation, infection and cancer (1). TNF-alpha triggers a signaling cascade, converging on the activation of the transcription factor NF-kappa B, which forms the basis for numerous physiological and pathological processes (2). TNF-alpha has been shown to play a role in systems such as the pathogenesis of psoriasis, tuberculosis, obesity, cerebral malaria, rheumatoid arthritis and asthma (3). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
