Tumor Necrosis Factor alpha (TNFa)

Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis. \n\nApplications: \nSuitable for use in ELISA, Western Blot, Neutralization and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nELISA: 1:200-1:1000 Best used as the second antibody in combination with a monoclonal antibody as a capture antibody.\nWestern Blot: 1:400-1:200 Supernatants or lysates of 2x10e6 endotoxin-stimulated human peripheral blood mononuclear cells (PBMC). PBMC are stimulated for 24 hours with 1%human serum plus 10 ng/mL E.coli LPS.\nImmunohistochemistry (Paraffin): 1:200\nNeutralization: 1:200 > 4hours\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes mature human TNFa at 17kD. Does not recognize human TNGb (lymphotoxin). \nRecognize the cell-bound precursor of TNFa as a 26kD protein in immunoblots, (denatured). Neutralizes human and primate TNFa activity Does not neutralize the biological activity of lymphotoxin.
