Tumor Necrosis Factor alpha (TNFa)

Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis. \n\nApplications: \nSuitable for use in RIA, ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nRIA: 1:8000 \nNeutralization: 1:200 (> 4 hours).\nImmunoprecipitation: 1:400-1:800 use non-specific rabbit IgG for pre-cleaning.\nELISA (soluble, extracellular): 1:1000-1:2000 IL-1RII using HRP Conjugated Goat Anti-Rabbit IgG (H&L) and ABTS Stable Liquid Substrate 1.46mM/Liter for HRPO A0030 as a substrate for 30 minutes at RT.\nImmunohistochemistry: 1:200\nWestern Blot: 1:400-1:800 IL-1RII using HRP Conjugated Goat Anti-Rabbit IgG (H&L) and TMB Stable Liquid Substrate for HRPO (3,3',5,5'-Tetramethylbenzidine) T6000 or T6005.\nOptimal dilutions to be determined by the researcher.\n\nRecommended Secondary Antibodies:\nI1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb\nI1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb\nI1904-46Q: IgG, H&L (HRP) Pab Gt xRb\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes the human and primate TNFa at 17kD and cell-bound precursor of TNFa as a 26kD protein in immunoblots, particularly in denatured samples. Does not recognize human TNGb (lymphotoxin).
