Tumor Necrosis Factor Receptor 2, p75/p80 (TNFR2, CD120b)

Tumor Necrosis Factor Receptor 2, p75/p80 (TNFR2) is present on most cell types and is considered to be an important co-stimulator together with TNF-RI in cellular responses to TNF-alpha, in particular the transmembrane form of TNF-alpha. A dominant role of TNFR2 has been shown in thymocyte activation by TNF-alpha. TNF-alpha activates inflammatory responses, induces apoptosis, regulates cellular proliferation, and may even promote cancer progression. The effects of TNF-alpha are mediated by TNFR1 and TNFR2, which have both distinct and overlapping downstream signalling cascades. Induction of cytotoxicity and other functions are mediated largely via TNFR1. TNFR1 is equally well activated by both the 17kD soluble and 26kD membrane-bound form, whereas TNFR2 is activated only by the membrane bound form of TNF-alpha. The antibody is a non-agonistic receptor modulating antibody. \n\nApplications:\nSuitable for use in Flow Cytometry, Immunofluorescence, Immunoprecipitation, ELISA and Functional Studes. Other applications have not been tested.\n\nRecommended Dilutions:\nFlow Cytometry: 1:50. Used to select HeLa cells. Mock transfected HeLa cells were used as a negative control (2).\nImmunofluorescence: 1:50\nFunctional Studies: Functions as an allosteric modulator which stabilizes the ligand-receptor complex (1).\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: HeLa cells transfected with TNFR2.\n\nNegative Control: Mock transfected HeLa cells\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
