Tumor Necrosis Factor Receptor 2 (TNFR2, CD120b)

TNF-alpha is an important cytokine produced by numerous cell types including neutrophils, activated lymphocytes, macrophages and NK cells. It plays a critical role in inflammatory responses and in apoptosis (1). TNF-alpha exists as a membrane-anchored and soluble form, both of which show biological activity. Response to TNF-alpha is mediated through two receptors, TNFR1, which is widely expressed, and TNFR2, which is expressed mainly in immune and endothelial cells (2). Antagonists to TNF-alpha have been validated as therapeutic targets for rheumatoid arthritis and other immune disorders (3). \nThe two receptors for TNF-alpha, TNFR1 (55kD) and TNFR2 (75kD) can mediate distinct cellular responses (4,5). In most cases cytotoxicity elicited by TNF has been reported to act throught TNFR1 (6,7). In contrast, TNFR2 appears to be important in T cell signaling and responses to infection (7,8). TNFR2 binds to distinct members of the TRAF family leading to the activation of NF-kappaB (9,10). Soluble forms of both receptors have also been characterized which can bind TNF-alpha and may play an important role in immune disorders.\n\nApplications: \nSuitable for use in Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1:1000\nImmunoprecipitation: 1:100\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
