Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with antibody showed tyrosinase in melanotic as well as amelanotic variants (1). This antibody is a useful marker for melanocytes and melanomas. \n\nApplications:\nSuitable for use in Immunocytology, Immunofluorescence, Western Blot and Immunohistology. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot: 1-2ug/ml for 2 hours at RT.\n\nImmunohistology: 2-4ug/ml for 30 minutes at RT (formalin/paraffin). Staining of formalin-fixed tissues requires boiling tissue sections in 1mM EDTA, pH 8.0, for 10-20 min followed by cooling at RT for 20 min.\n\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nMelanoma cell lines (SK-MEL-13, SK-MEL-19, SK-MEL-30, SK-MEL-37), melanocytes in skin and Melanomas (1)\n\nCellular Localization: \nCytoplasmic\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C or colder. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes a cluster of proteins between 70-80kD, identified as tyrosinase. Occasionally a minor band at 55kD is also detected. Does not crossreact with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Species Crossreactivity: Human, canine and feline. Weak crossreactivity with mouse.
