Tyrosinase (TYR, LB24 AB, LB24-AB, Monophenol Monooxygenase, Oculocutaneous Albinism IA, OCA1A, OCAIA, SK29 AB, SK29-AB, Tumor Rejection Antigen AB)

Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy- phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with antibody showed tyrosinase in melanotic as well as amelanotic variants (1). This antibody is a useful marker for melanocytes and melanomas. \n\nApplications:\nSuitable for use in Immunocytochemistry, Western Blot, Immunofluorescence and Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilutions:\nWestern Blot: 1-2ug/ml for 2 hours at RT.\nImmunohistochemistry (formalin/paraffin): 2-4ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 1mM EDTA, pH 8.0, for 10-20 minutes followed by cooling at RT for 20 minutes.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nMelanoma cell lines and Melanoma\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes a cluster of proteins between 70-80kD, identified as human tyrosinase. Occasionally a minor band at 55kD is also detected. Does not crossreact with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Species Crossreactivity: canine and feline. Weak crossreactivity with mouse.
