Tyrosine Hydroxylase (TH, TYH, Tyrosine 3 Hydroxylase, Tyrosine 3 Monooxygenase)

Tyrosine hydroxylase (TH) plays an important role in the physiology of adrenergic neurons. It is the first and rate-limiting enzyme involved in the biosynthesis of the catecholamines Dopamine and Norepinephrine from tyrosine. TH is, therefore, a useful marker for dopaminergic and noradrenergic neurons. The enzymatic activity of TH requires ferrous ions as cofactors and is believed to be regulated by phosphorylation. At least four isoforms of human TH have been identified which result from alternative splicing.\n\nApplications:\nSuitable for use in Western Blot, ELISA, Immunohistochemistry, Immunofluorescence and Immunoprecipitation. Other applications have not been tested.\n\nRecommended Dilutions:\nWestern Bot: 1:1000 on 10ug of PC12 lysates.\nImmunohistochemistry: 1:100-1:1000 at 4 degrees C, 1-3 days. Stains both fresh frozen and paraffin embedded tissue samples by indirect immunofluorescence and immunoperoxidase. Not suitable for use on paraffin embedded, human tissue.\nImmunofluorescence: 1:100-1:1000; Fixed with 4% PFA overnight at 4 degrees C.\nOptimal dilutions to be determined by the researcher.\n\nRecommended Secondary Antibodies:\nI1904-39: IgG, X-Adsorbed (HRP) Pab Gt x Rb\nI1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt x Rb\nI1904-46Q: IgG, H&L (HRP) Pab Gt x Rb\n\nPositive Control: Brain (corpus striatum, sympathetic nerve terminals) and adrenal glands\n\nNegative Control: Liver\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
