Tyrosine Hydroxylase (TH, TYH, Tyrosine 3 Hydroxylase, Tyrosine 3 Monooxygenase)

Tyrosine hydroxylase plays an important role in the physiology of adrenergic neurons. It is the first and rate-limiting enzyme involved in the biosynthesis of the catecholamines Dopamine and Norepinephrine from tyrosine. TH is, therefore, a useful marker for dopaminergic and noradrenergic neurons. The enzymatic activity of TH requires ferrous ions as cofactors and is believed to be regulated by phosphorylation. At least four isoforms of human TH have been identified which result from alternative splicing.\n\nApplications: \nSuitable for use in Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1:1000 on 10ug of mouse brain lysates\nImmunohistochemistry (Frozen): 1:200-1:400, 4% PFA fixed, frozen sections\nImmunohistochemistry (PFA fixed, paraffin): 1:100 (Barrachina, M. et al., 2003). For paraffin sections Barrachina reported successful staining with microwave citrate acid antigen recovery, however other methods can likely be used as well.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
