UBCH9 (Ubiquitin-conjugating Enzyme E2 E3, Ubiquitin-protein Ligase E3, Ubiquitin Carrier Protein E3, Ubiquitin-conjugating Enzyme E2-23kD, UbcH9, UBE2E3, UBCE4)

SUMO conjugation to target proteins is mediated by a different, but analogous, pathway to ubiquitinylation. UbcH9 is a SUMO conjugating enzyme that mediates the conjugation of SUMO-1 to a number of proteins including RanGAP1, IkBa and PML. It is known to interact directly with proteins that are modified by SUMOylation and may be important for substrate recognition. It appears that conjugation of SUMO-1 to IkBb prevents subsequent ubiquitinylation and thus protects it from proteasomal degradation. Thus, while ubiquitinylation targets proteins for rapid degradation, SUMO-1 modification acts antagonistically to generate proteins resistant to degradation.\n\nApplications:\nSuitable for use in Western Blot and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 0.5ug/ml\nImmunocytochemistry: 1ug/ml\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nNIH/3T3 cell lysate\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
