The E2 enzyme, Ubc13, and the E2 enzyme variants, Uevs, form stable, high affinity complexes for the assembly of Lys63-linked ubiquitin chains. This complex has been shown to activate NF-kB signaling pathway by linking TRAF6 to IKKs. Blockade of this polyubiquitin chain synthesis, but not inhibition of the proteasome, prevents the activation of IKK by TRAF6. These results suggest a new regulatory function for ubiquitin, in which IKK is activated through the assembly of Lys63-linked polyubiquitin chains. The UEV proteins are also involved in the control of differentiation. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1-3ug/ml Optimal dilutions to be determined by the researcher. Positive Control: Jurkat, HeLa or RAW (mouse) cell lysates. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
