UNC5CL is a membrane protein belonging to theUNC-5 family with a death domain and a ZU5 domain and is known to interact with p65:RELA and NFKB1. UNC5CL is known to interact with subunit p105 of NF-kappaB and transactivator p65 and is thus involved in inhibition of NF-kappaB activation and NF-kappaB-dependent transcription by inhibiting the binding of NF-kappaB to its target sequence (providing an alternative regulatory mechanism for NF-kappaB-mediated transcription). This regulatory protein sensitizes cells to apopotosis induced by TNF and TRAIL.
