Human vimentin is a 53,521 dalton protein (466aa) expressed in many cell types of mesodermal origin, and is one of the intermediate filament types that form the cytoskeleton. In the embryonic nervous system, however, vimentin is expressed by migrating neural crest cells forming the PNS, and by neural stem cells of the CNS. In some forms of glioblastoma, vimentin is re-expressed by the tumor cells and serves as a general indicator of their undifferentiated state. Immunohistochemical staining for Vimentin is also characteristic of sarcomas (of neural, muscle and fibroblast origin) compared to carcinomas which are generally negative. Melanomas, lymphomas and vascular tumors may all stain for Vimentin. Vimentin antibodies are thus of value in the differential diagnosis of undifferentiated neoplasms and malignant tumors. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:250-1:500 Immunohistochemistry: 1:100-1:200 Immunocytochemistry: 1:100-1:200 IOptimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1906: IgY, Chicken Pab Rb x Ch I1906-10: IgY, Chicken (HRP) Rb x Ch I1906-12: IgY, Chicken (FITC) Rb x Ch I1906-14: IgY, Chicken (AP) Rb x Ch Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
