VISA is an important protein required for innate immune defence against viruses. VISA acts downstream of DDX58 and IFIH1/MDA5, which detect intracellular dsRNA produced during viral replication, to coordinate pathways leading to the activation of NF-kappa-B, IRF3 and IRF7, and to the subsequent induction of antiviral cytokines such as IFN-beta and RANTES (CCL5). VISA is believed to activate the same pathways following detection of extracellular dsRNA by TLR3 and it has been hypothosized that VISA may protect cells from apoptosis. VISA is a ubiquitously expressed protein present in a range of cell types including T-cells, monocytes and epithelial cells, highest levels in tissues of the heart, skeletal muscle, liver, placenta and in peripheral blood leukocytes. Multiple isoforms of VISA have been identified. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5-1ug/ml Immunohistochemistry (Paraffin): 2.5ug/ml Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: Sheep Anti Rabbit IgG HRP Immunohistochemistry Positive Control Tissue: Mouse brain Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
