Wee1

Wee1 is one of the nuclear tyrosine protein kinases which participates in the regulation of multiple cellular processes including transcription, DNA repair and the cell cycle. Structurally this kinase belongs the serine/threonine PK family but it possesses tyrosine kinase actiivity (1). Human Wee1 phosphory-lates Y15 of Cdc2 and ensures the completion of DNA replication prior to mitosis by inhibiting the activity of the Cdc2-cyclin B kinase during the S and G2 phases of the cell cycle. Cdc2-cyclin B kinase is required for the pro-gression of cells into M phase. However, phosphorylation of T14 or Y15 residues of Cdc2 inhibits its activity (2).\n\nApplications: \nSuitable for use in Western Blot. Other applications not tested. \n\nRecommended Dilution:\nWestern Blot: A 1:500-1:2000 detected Wee1 in RIPA lysates from unstimulated A431 cells.\nOptimal dilutions to be determined by the researcher.\n \nIncluded Positive Antigen Control: A0001-50\nAdd 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
