Wilms Tumor (WT) is a 55kDa protein thought to play a major role in various cancers and developmental disorders, commonly kidney tumors and glomerular diseases. WT is a zinc finger transcription factor that is mutated in Wilms tumors and highly expressed in a wide variety of other malignancies (1). It is believed that WT in its two isoforms acts as both a transcription factor and a post-transcriptional regulator that binds RNA and can interact with splicing components (2). Mutations in the zinc finger regions of WT are detected in nearly all patients of Denys-Drash syndrome and in some patients with isolated diffuse mesangial sclerosis. Also, mutations leading to the loss of the transcription factor isoform have been observed in all patients with Frasier syndrome (3). Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilution: Western Blot: 1:1000-1:5000 Immunohistochemistry: 1:100- 1:250 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
