Wnt3a (Wingless-type MMTV (Mouse Mammary Tumor Virus) Integration Site Family Member)

The Wnt gene family consists of structurally related genes, which encode secreted signaling proteins.1 Wnt proteins have been implicated in oncogenesis 2-3 and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. The mouse studies show the requirement of Wnt3 in primary axis formation in the mouse. Studies of the gene expression suggest that this gene may play a key role in some cases of human breast, rectal, lung, and gastric cancer through activation of the Wnt-beta-catenin-TCF signaling pathway.3\n\nApplications: \nSuitable for use in ELISA and Western Blot. Other applications not tested.\n\nRecommended Dilution:\nELISA: 0.5-2ug/ml detects hWnt-3a by indirect ELISA using 100ul/well antibody solution. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2-0.4ng/well of recombinant hWnt-3a.\n\nWestern Blot: 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hWnt-3a is 1.5-3ng/lane, under either reducing or non-reducing conditions.\n\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nHuman Wnt3 transfected 293 cells\n\nStorage and Stability:\nLyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20 degrees C. Reconstituted product is stable for 24 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
