ZAP 70 (TCR Zeta Associated Protein kinase 70kD)

Zap-70, a Syk family protein tyrosine kinase expressed in T and NK cells, plays a critical role in mediating T cell activation in response to T cell receptor (TCR) engagement (1). Following TCR engagement, Zap-70 is rapidly phosphorylated on several tyrosine residues, presumably by two mechanisms: autophosphorylation and transphosphorylation by the Src family tyrosine kinase, Lck (2-6). Tyrosine phosphorylation of Zap-70 correlates with its increased kinase activity and downstream signaling events. In patients with chronic lymphocytic leukemia (CLL), total Zap-70 expression was shown to be correlated with disease progression and survival (7,8).\n\nApplications: \nSuitable for use in Flow Cytometry, Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution:\nFlow Cytometry: 1:200\nWestern Blot: 1:2000\nImmunoprecipitation: 1:50\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
