The polymerase chain reaction (PCR), in which a minute amount of DNA molecules are copied, or amplified, is foundational to biotechnology, clinical testing, molecular diagnostics, drug discovery and development, food & beverage testing, forensics, genomics, plant biology, proteomics, stem cell technologies and many others.

The PCR reaction begins when the double strands of a DNA molecule are separated by heating. Then, after cooling the sample down, short strands of two specific DNA primers are able to anneal, or bind to either end of the target stretch of code. Next, the temperature is raised to a midpoint and Taq polymerase is added which binds to both of the primers and synthesizes a complementary copy of the DNA sequence using nucleotides present in the solution. This process can be repeated, each time doubling the amount of DNA present, until millions of copies of the target sequence have been synthesized in a few hours.

In order to monitor the PCR reaction and determine the concentration of target molecules created, fluorescent signals from probes bound to sequences of DNA can be optically detected and recorded.
