4-1BB Ligand (TNFSF9, CD137, CDw137, ILA, MGC2172, T-cell, 4-1BB Homolog, T-cell Antigen ILA, Tumor Necrosis Factor Receptor Superfamily Member 9 Precursor)

4-1BB, also known as CD137, is a type I transmembrane protein belonging to the TNF receptor superfamily (TNFRSF9). It is expressed on T cells, NK cells, monocytes, neutrophils, dendritic cells, and eosinophils. The ligand for 4-1BB, 4-1BBL, is a TNF ligand superfamily member (TNFSF9) expressed on the surfaces of activated antigen presenting cells and T and B lymphoma cell lines. 4-1BB transduces a T cell costimulatory signal to promote survival, proliferation, cytokine production, and effector functions. On dendritic cells, 4-1BB ligation enhances cytokine production and upregulates expression of B7-1 and B7-2 costimulatory molecules. \n\nApplications:\nSuitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nELISA: To detect h4-1BBL by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.5-2ug/ml of this antibody is required.\nWestern Blot: To detect h4-1BBL by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. When used in conjunction with compatible secondary reagents, the detection limit for recombinant h4-1BBL is 1.5-3ng/lane, under either reducing or non-reducing conditions.\nImmunohistochemistry (Formalin fixed paraffin embedded): This antibody stained formalin-fixed, paraffin-embedded sections of human colon/rectum adenocarcinoma. The recommended concentration is 0.25 mg/ml with an overnight incubation at 4 degrees C. An HRP-labeled polymer detection system was used with a DAB chromogen. Proteinase K antigen retrieval is recommended.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
