Alpha adducin is a 80-120kD member of the aldolase class II family and adducin subfamily. Adducin is a membrane skeletal protein that binds to actin filaments (F-actin) and promotes the association of spectrin with F-actin to form a spectrin-actin meshwork beneath plasma membranes (1-2). Adducin is a heterodimeric protein that consists of related subunits; a and b, or a and g subunits (3). Adducin binds with high affinity to Ca(2+)/calmodulin and is a substrate for protein kinase C (PKC), and protein kinase A (PKA) (4). PKA specifically phosphorylates Ser59, Ser408, Ser436, and Ser481 located in the neck domain of alpha Adducin. Phosphorylation of Adducin alpha by Rho-kinase at Thr445 and Thr480 enhances the F-actin-binding activity of Adducin alpha (5). Applications: Suitable for use in Western Blot, Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:2000-5000 Immunoprecipitation: 1:40 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Manufactured incorporating RabMAb(R) technology under Epitomics US patents, No 5,675,063 and 7,429,487, owned by Abcam.
