b-Agarase is used to purify nucleic acids and DNA fragments from low melting agarose gels by hydrolyzing the polysaccharide core of 1,3-b,D-galactopyranose into neoagarobiose oligosaccharides. Unit Definition: One unit of b-Agarase is defined as the amount of enzyme necessary to produce 1ug of reducing sugar as D-Galactose from agar at pH 6.0 at 40 degrees C per minute. Activity: ~1000-3000U/mg Application: Quantitative recovery of DNA or RNA from low melting point agarose gels. The recovered nucleic acids can be subjected to many further manipulations such as restriction endonuclease digestion, ligation, labeling, sequencing, amplification, etc. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
