APG4 is a cysteine protease required for autophagy which cleaves the C-terminal part of either MAP1LC3, GABARAPL2 or GABARAP, allowing the liberation of form I. A subpopulation of form I is subsequently converted to a smaller form (form II). Form II, with a revealed C-terminal glycine, is considered to be the phosphatidylethanolamine (PE)-conjugated form, and has the capacity for the binding to autophagosomes. The protein is highly expressed in human skeletal muscle, heart, liver and testis. The protein is highly conserved from yeast to higher eukaryotes. Alternate transcriptional splice variants, encoding the same protein, resulting in two isoforms (isoform 7:8), have been characterized. The gene encoding for human APG4C Isoform 7:8 is localized on chromosome 1p31.3. Applications: Suitable for use in ELISA. Other applications not tested. Recommended Dilution: ELISA: 1:100-1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
