Catenin, beta, phosphorylated (Tyr142)

b-Catenin is a 92kD protein that binds to the cytoplasmic tail of E-Cadherin. The cadherins, transmembrane adhesion molecules, are found with catenins at adherens junctions. Deletions in the cytoplasmic domain of E-Cadherin eliminate catenin binding and result in a loss of cell adhesion. Tyrosine phosphorylation of b-Catenin can regulate its interaction with critical components of adherens junctions. Both Fer and Fyn kinases phosphorylate tyrosine 142 in vitro. Overexpression of these kinases in epithelial cells disrupts interactions between a- and b-Catenins. The phosphorylation of tyrosine 142 may act as a switch from the transcriptional to the adhesive role of b-Catenin. Src family kinases can also phosphorylate tyrosine 654 in the C-terminal armadillo repeat of b-Catenin. This phosphorylation regulates b-Catenin binding to E-cadherin. Thus, site-specific tyrosine phosphorylation of b-Catenin may regulate specific protein-protein interactions leading to changes in cell adhesion.\n\nApplications: \nSuitable for use in ELISA, Western Blot or for antigen applications in immunological protocols. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:500. Membrane was incubated with diluted antibody in 5% non-fat milk, PBS, 0.04% Tween 20 for 1 hour at RT.\nELISA: 1:2000\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes a 92kD protein corresponding to the molecular mass of b-Catenin wnen phosphorylated at Try142 on SDS-PAGE immunoblots of Hct116 src transformed cells treated with pervanadate, but not in control cells. Similar results were observed in other pervanadate-treated cell lines, such as A431 and human endothelial cells. Species sequence homology: mouse and rat. Sequence homology: one amino acid difference from a sequence around tyrosine 133 of human g-Catenin.
