CD235a (Glycophorin A, GpA) (Biotin)

Human CD235a (glycophorin A) is a sialoglycoprotein expressed early on the cell surface during erythropoiesis as pluripotent stem cells are recruited to differentiate into erythrocytes (1). CD235a (glycophorin A) is a useful marker to study the developmental biology of hematopoietic progenitor cells (2,3).\n\nApplications: \nSuitable for use in Flow Cytometry. Other applications have not been tested. \n\nRecommended Dilutions:\nFlow Cytometry: 10ug/ml\nOptimal dilutions to be determined by the researcher.\n\nFlow Cytometry Protocol:\n1. 2ul per tube of freshly drawn heparinized human whole blood was washed and incubated for 45 min on ice with 80ul of C2548-98D2 at 10ug/ml. \n2. Cells were washed twice and incubated with S7973-88P, Streptavidin/R-Phycoerythrin, after which they were washed three times and fixed.\n3. To avoid clots, fixed red blood cells were vortexed and drip-filtered through a 35um nylon mesh filter prior to analysis by FACS. \n4. Cells stained positive with a mean shift of 1.34 log10 fluorescent units when compared to a mouse IgM/Biotin negative control at a similar concentration. \n5. Binding was blocked when cells were pre-incubated for 10 minutes with 20ul of 0.5mg/ml of C2548-98D1 CD235a (Glycophorin A, GpA).\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
