CD33 antigen is a useful marker for the diagnosis of non lymphoblastic leukemias and also for distinguishing myelogenous leukemia cells from lymphoid leukemia cells. The CD33 antigen is earlier expressed in the cytoplasm (cyCD33) than the expression on the cell membrane (mCD33) of the myeloblasts. It is more valuable using cyCD33 and cyCD13 to diagnose leukemia of AML-M0 (as the CD13 antigen is also earlier expressed in the cytoplasm (cyCD13) than the expression on the cell membrane (mCD13) of the myeloblasts). The CD33 antigen functions as an adhesion molecule. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (Frozen): 1:50-1:100. Overnight incubation recommended. Not recommended for use with paraffin sections. Flow cytometry: 1:10-1:50; 10ul labels 10e6 cells Optimal dilutions to be determined by the researcher. Immunohistochemistry Positive Control: Lymphatic tissue Recommended Negative Control: I1904-78R:IgG1 Murine Negative Control Hybridoma: NS1 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
