CD74 (HLA-DR, DHLAG, HLA class II histocompatibility antigen gamma, HLADG, Ia antigen-associated invariant chain, Ia-Gamma, Ii, p33, protein 41 antigen invariant chain)

CD74, also known as the MHC class II-associated invariant chain (Ii), is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevent their premature association with endogenous polypeptides. CD74 is produced in molar excess over MHC class II and some of this is expressed by an unknown pathway on the cell surface independent of or in association with MHC class II molecules (1,2). The half-life of CD74 on the cell surface is only 3-4 min after which it is internalized. It has been proposed that cell surface CD74 binds MHC class II molecules which have lost their antigenic peptide leading to internalization and reloading with antigenic peptide (3).\n\nApplications:\nSuitable for use in Western Blot, Immunoprecipitation, Immunocytochemistry and Flow Cytometry. Other applications not tested. \n\nRecommended Dilution: \nWestern Blot (ECL and colorimetric): 1ug/ml\nImmunoprecipitation: 12ug/ml\nOptimal dilutions to be determined by researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
