COX 2 (Cyclooxygenase 2, PGHS 2, Prostaglandin Endoperoxide Synthase 2, PHS 2)

Cyclooxygenases 1 and 2 (COX-1, COX-2) are enzymes involved in the conversion of arichidonate to prostaglandins. COX-1 and COX-2 are very similar in structure and function, though they vary in expression. COX-1 is normally expressed in most cell types, whereas COX-2 expression is at low levels unless induced by hormonal stimuli. The apparent molecular weight of COX-2 appears to be around 70kD. \nCOX-2 is the inducible isoform of cyclooxygenase. COX-2 is upregulated in response to number of mediators, including growth factors, cytokines and endotoxins. COX-2 appears to be expressed in several different types of human cancers, including sporadic coloreactal, breast, pancreatic and cervical cancers. COX-2 immunoreactivity is also present in neurons at postsynaptic sites in rat cerebral cortex. Besides its role in carcinogenesis, COX-2 is also involved in apoptosis, cellular adhesion as well as angiogenesis.\n\nApplications:\nSuitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilution:\nELISA: 0.1-1ug/ml\nWestern Blot: 1-3ug/ml\nImmunohistochemistry: 10-20ug/ml. Formalin-fixed, paraffin-embedded tissue sections require heat induced epitope retrieval (HIER) with citrate buffer, pH 6.0, prior to staining.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
