Cytokeratin 5,6

The cytokeratins are a family of intermediate filaments fond in almost all epithelial cells. They are divided into four subclasses depending on their patterns of reactivity and electrophoretic characteristics and are also arranged I a numbered series of 20 or more acoring to molecular weight and isoelectric point (Quinlan et al. 1985). Types 5 and 6 of cytokeratin (CK5/6) are in high frequency in mesothelial cells, as opposed to cells of adenocarcinoma. CK5/6 has been shown to be a reliable marker for mesothelioma and squamous cell carcinoma of the lung and Bowen's disease. It does not react with pulmonary adenocarcinoma.\n\nApplications: \nSuitable for use in Immunohistochemistry, Western Blot, Immunofluorescence. Other applications not tested.\n\nRecommended Dilution: \nImmunohistochemistry (frozen or paraffin sections): 1:50-1:100\nWestern Blot: Recognizes a 58kD and 56kD band corresponding to cytokeratin 5 and 6. Cytoskeletal preparations of epidermis.\nOptimal dilution determined by the researcher. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes human cytokeratin 5. Demonstrates crossreactivity with cytokeratin 6. Demonstrates weak crossreactivity with cytokeratin 4 on immunoblots. Does not crossreact with cytokeratins 1, 7, 8, 10, 13, 14, 18 or 19. Recognizes basal cells and a part of stratum spinosum in the normal pavement epithelium.
