Desmocollin 3

Desmocollin 3 is a calcium-dependent glycoprotein that is a member of the desmocollin subfamily of the cadherin superfamily. These desmosomal family members, along with the desmogleins, are found primarily in epithelial cells where they constitute the adhesive proteins of the desmosome cell-cell junction and are required for cell adhesion and desmosome formation. The desmosomal family members are arranged in two clusters on chromosome 18, occupying less than 650kb combined.\nDsc3 is present in all living epidermal layers, in glandular duct cells, in basal matrix cells and outer root sheath of hair follicles, basal and suprabasal cell layers of stratified epithelia (e.g. vagina, tongue, esophagus), basal layer of bladder urothelium and complex epithelium of trachea, and thymic reticulum cells. \nAll simple epithelia are negative. Intercalated disks of myocardium are negative. Polypeptide reacting: 109kD and 10kD polypeptides (desmocollin 3 splice isoforms at pI 5.2) from human epidermal desmosomes. Most cultured cell lines derived from stratified squamous epithelia or squamous cell carcinomas are positive (e.g. A-431, HaCaT).\n\nApplications:\nSuitable for use in Immunohistochemistry and Western Blot. Other applications not tested. \n\nRecommended Dilutions: \nImmunohistochemistry: 1:10 in PBS, pH 7.4. Incubation Time 1 h at RT. Frozen and paraffin-embedded sections after microwave treatment. For better resolution on frozen sections preincubation (directly after fixation) with 0.05-0.2% Triton X-100, for 5-10 minutes, depending on tissue type, is recommended.\nOptimal dilutions to be determined by the researcher. \n\nStorage and Stability: \nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
