DFF (DNA Fragmentation Factor)

Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. These death signals finally cause the degradation of chromosomal DNA by activated DNase. A human DNA fragmentation factor (DFF) was identified recently which was cleaved by caspase-3 during apoptosis(1). A mouse homologue of human DFF was identified as a DNase inhibitor and designated ICAD(2,3). Upon cleavage of DFF/ICAD, a caspase activated deoxyribonuclease (CAD) is released and activated and eventually causes the degradation of DNA in the nuclei(2,3). Therefore, the cleavage of CAD inhibitor molecule DFF/ICAD, which causes DNase activation and DNA degradation, is the hallmark of apoptotic cell death(4). \n\nApplications: \nSuitable for the detection of DFF by Western blot and for immuno-precipitation and coprecipitation to analyze DFF interacting proteins. Whole cell lysate from HeLa cells can be used as positive control and a 45kD band is detected. Western blot analysis of 50ug of total cell lysate from HeLa cells with anti-DFF (CT) at 1:2000 dilution. \n\nRecommended Dilution: \nWestern blot: 1:1000 to 1:2000\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
