FAK (Focal Adhesion Associated Protein Tyrosine Kinase, BC3)

FAK is a widely expressed non-receptor protein tyrosine kinase (PTK). FAK has a central catalytic domain and a C-terminal tail that localizes it to focal adhesions, which are sites where cells attach to the extracellular matrix via surface integrin receptors. Increased FAK tyrosine phosphorylation occurs upon integrin engagement with fibronectin. Studies have shown that adhesion of murine NIH3T3 fibroblasts to fibronectin promotes association of the Grb2 adapter protein and c-Src PTK with FAK in vivo, and also results in activation of the ERK2 MAP kinase. In v-Src-transformed NIH3T3, the association of v-Src, Grb2, and Sos with FAK is independent of cell adhesion to fibronectin. In vitro the Grb2 SH2 domain binds directly to tyrosine- phosphorylated FAK, and the binding site has been identified as Tyr925 by site directed mutagenesis. \n\nApplications: \nSuitable for use in Immunoprecipitation and Immunofluorescence. Not recommended for Western Blot.\nOther applications have not been tested.\n\nRecommended Dilution:\nImmunofluorescence: 10ug/ml\nImmunoprecipitation/Western Blot: 4ug immunoprecipitates FAK from 500ug of a mouse 3T3/A31 cell RIPA lysate. The immunoprecipitate was resolved by electrophoresis, transferred to nitrocellulose and probed with F0019-56 (2ug/ml). Proteins were visualized using an IgG (HRP) goat anti-rabbit secondary antibody and a chemiluminescence detection system. \nOptimal dilutions to be determined by the researcher.\n\nRecommended Secondary Antibodies:\nI1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb\nI1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb\nI1904-46Q: IgG, H&L (HRP) Pab Gt xRb\n\nStorage and Stability:\nFor long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
