FAK (Focal Adhesion Associated Protein Tyrosine Kinase, BC3)

FAK is a widely expressed non-receptor protein tyrosine kinase (PTK). FAK has a central catalytic domain and a C-terminal tail that localizes it to focal adhesions, which are sites where cells attach to the extracellular matrix via surface integrin receptors. Increased FAK tyrosine phosphorylation occurs upon integrin engagement with fibronectin. Studies have shown that adhesion of murine NIH3T3 fibroblasts to fibronectin promotes association of the Grb2 adapter protein and c-Src PTK with FAK in vivo, and also results in activation of the ERK2 MAP kinase. In v-Src-transformed NIH3T3, the association of v-Src, Grb2, and Sos with FAK is independent of cell adhesion to fibronectin. In vitro the Grb2 SH2 domain binds directly to tyrosine- phosphorylated FAK, and the binding site has been identified as Tyr925 by site directed mutagenesis. \n\nApplications: \nSuitable for use in Immunohistochemistry, Western Blot, Immunocytochemistry, Immunoprecipitation and ELISA. Other applications not tested. \nRecommended Dilutions: \nWestern blot: 0.2-1ug/ml of this lot detected FAK from a 3T3/A31 RIPA cell lysate. 0.2-1ug/ml detected FAK from PC-12, Hep-G2, SW 620 and Jurkat RIPA cell lysates. 3T3/A31 cell lysate was resolved by electrophoresis, transferred to PVDF and probed with F0019-55A (0.2ug/ml). Proteins were visualized using a IgG (HRP) goat anti-mouse and a chemiluminescence detection system.\nImmunocytochemistry, Immunohistochemistry: 2.5-5ug/ml (human breast carcinoma cells).\nImmunoprecipitation: 5ug immunoprecipitated FAK from 250ug of 3T3/A31 RIPA cell lysate. \nOptimal dilutions to be determined by the researcher.\n\nIncluded Positive Antigen Control:\nF4209 Fibroblast Cell, 3T3/A31\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
