Fgr, active, Recombinant, Human (SRC2, c-FGR, p55 c-fgr, Human Gardner-Rasheed feline viral oncogene)

FGR is a proto-oncogene and a unique member of the tyrosine kinase family (1). Certain lymphomas (but not sarcomas or carcinomas) express FGR-related mRNA. This transcript is detected in Burkitt's lymphoma cell lines naturally infected with Epstein-Barr virus (EBV), but not in EBV-negative Burkitt's lymphoma cells (2). Normal umbilical cord or peripheral blood lymphocyte lines established in vitro by EBV infection also contain detectable c-FGR mRNA. Moreover, a 50-fold increase of the steady-state c-FGR mRNA concentration is observed when \nuninfected Burkitt's lymphoma cell lines are deliberately infected with EBV demonstrating the induction of a proto-oncogene in response to infection by a DNA tumor virus. FGR expression is limited to normal peripheral blood granulocytes, monocytes, and alveolar macrophages, all \nof which contain 50 to 100 copies of c-FGR mRNA per cell (3). Very low levels of FGR transcripts are also present in U937 promonocytic cells and increase in abundance with 12-O-tetradecanoylphorbol-13-acetate (TPA)- induced differentiation. Cycloheximide also causes accumulation of c- FGR transcripts in U937 cells. \n\nSpecific Activity: 374nmol/min/mg\n \nActivity Assay: \nKinase activity is measured as the molar amount of phosphate incorporated into Poly(Glu-Tyr) substrate per minute per mg protein at 30 degrees C using a final concentration of 50uM [32P] ATP.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot to avoid repeated freezing and thawing and freeze at -70 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 12 months.
