Fibroblast Growth Factor 2, Basic, Neutralizing (FGF2b) ( BSA, Azide, Glycerol Free)

bFGF (FGF-2) is a member of a family of seven peptides that are potent regulators of cell proliferation, differentiation and function. bFGF stimulates the proliferation of cells of mesodermal origin and many cells of endodermal, ectodermal and neuroectodermal origin. bFGF also induces neuron differentiation, survival and regeneration and stimulates astrocyte migration. bFGF may also be involved in the production of a variety of pathological conditions arising from excessive cell proliferation.\n\nApplications: \nSuitable for use in Neutralization. Not suitable for Western Blot, use F4210-21 Fibroblast Growth Factor 2, Basic (FGF2b). Other applications have not been tested.\n\nRecommended Dilution:\nNeutralization of bFGF Activity Assay: 2.5ug/ml\nNeutralizes >40% growth of bovine capillary endothelial cells simulated with 5ng/ml of FGF-2/bFGF. \nOptimal dilutions to be determined by the researcher.\n\nPositive Control: fetal liver or kidney tissue.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. \n\nNeutralization Assay Protocol \n1. Prepare a series of cell suspensions (4.5ml) of 10e4 cells/ml in RD (RPMI-1640/DMEM [1:1]) medium containing 2% FBS and 0.1ug/ml heparin. \n2. Incubate 5ng bFGF and different concentrations of F4210-22 in 4.5ml assay media, for 1 hour at 37 degrees C. \n3. Spin cells and resuspend in pre-incubated bFGF/a-bFGF (4.5ml) from step 2. Seed 1ml per well. \n4. On day 3, refeed each well with 1ml freshly prepared bFGF/a-bFGF immunocomplex as described in step 2. \n5. Count and record cell numbers on day 5.
