This Human Fibroblast Growth Factor basic (FGF-b) ELISA kit is to be used for the quantitative determination of FGF basic in human serum, plasma, buffered solution, or cell culture medium. The assay will recognize both the natural 18kD isoform and recombinant FGF basic-146 and FGF basic-157.\n\nSensitivity: \n<7pg/ml\n\nRange: \n15.6-1000pg/ml\n\nIncubation Time: \n4 hours, RT\n\nSpecificity: \nRecognizes the natural 18kD form of FGFb, as well as the recombinant FGFb-146 and recombinant FGFb-157.\n\nSimple: \nPre-coated strip-well plates. Heparin and citrate plasma are not recommended.\n\nPrinciple of the Method:\nThis human FGF basic kit is a solid phase sandwich ELISA Assay. A monoclonal antibody specific for human FGF basic has been coated onto the wells of the microtiter strips provided. Samples, including standards of known human FGF-b content, control specimens, and unknowns, are pipetted into these wells. During the first incubation, the FGF-b antigen binds to the immobilized (capture) antibody on one site. After washing, a biotinylated monoclonal antibody specific for FGF-b is added. During the second incubation, this antibody binds to the immobilized FGF-b captured during the first incubation. After removal of excess second antibody, Streptavidin-Peroxidase (enzyme) is added. This binds to the biotinylated antibody to complete the four-member sandwich. After a third incubation and washing to remove all the unbound enzyme, a substrate solution is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of FGF-b present in the original specimen.\n\nKit Components:\nF4210-19A: Microtiter Plate, 1x96wells. Contains 3.3mM thymol.\nF4210-19G: Wash Buffer Concentrate, 25X, 1x100ml.\nF4210-19H: TMB, 1x25ml.\nF4210-19J: Stop Solution, 1x25ml.\n\nStorage and Stability:\nStore at 4 degrees C. Store F4210-19B powder at 4 degrees C, as liquid at -70 degrees C. Stable for 6 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
