Fibronectin exists in three forms: 1. Soluble dimeric form, plasma fibronectin 2. Oligomers which are transiently attached to the cell surface, cell-surface fibronectin 3. Highly insoluble fibrils in the extracellular matrix, matrix fibronectin.\n\nHuman fibronectin is an adhesive glycoprotein with a molecular weight of 500kD found in the extracellular matrix and in plasma. Fibronectin is composed of two nearly identical polypeptide chains linked by a pair of disulfide bonds. Each subunit is organized into a variety of structural domains containing specific binding sites for cells, collagen, fibrin and glycosaminoglycans. Fibronectin also participates in complex interactions with other macromolecules to influence cellular properties including adhesion, morphology, migration, hemostasis and oncogenic transformation. The human fibronectin provided is suitable to promote the adhesion and propagation of cells in vitro when used to coat cell culture surfaces, including plasticware, glassware and microcarrier beads.\n\nCellular fibronectin functions in mesenchymal cell attachment embryogenesis, wound repair and cellular metastasis, a different function from plasma fibronectin.\n\nSource:\nFibronectin, from human plasma\n\nBiological Activity: \nCorning 24-well plates were coated with fibronectin in PBS (0.3ml/well) at 37 degrees C overnight. After blocking by BSA, TIG-3 cells (5 x 104 cells/1ml DMEM/well) were added and incubated at 37 degrees C for 90 minutes. Attached cells were counted electronically with a Coulter counter. Under these conditions, the use of a concentration of 6ug fibronectin per milliliter resulted in the attachment of greater than 70% of the cells.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
