FK506 Binding Protein 8 (FKBP8, FK506 Binding Protein 38kD, FKBP38, Rotamase)

FK506 Binding Proteins (FKBPs) are intracellular receptors for the immuno-suppressive drug FK506. The FKBP/FK506 complex exerts its immunosuppressive effects by inhibiting calcineurin, a calcium- and calmodulin-dependent serine/threonine phosphatase that functions as a critical signaling molecule during T-cell activation. FKBP38, also known as FKBP8, is a 355aa protein with a calculated molecular weight of 38.7kD and an apparent molecular mass of ~60-64kD in SDS-PAGE. FKBP38 binds to and inhibits calcineurin even in the absence of FK506, indicating that FKBP38 is a constitutively active inhibitor of calcineurin. Additionally, FKBP38 co-immunoprecipitates with Bcl-2 and Bcl-xL, suggesting that FKBP38 may regulate apoptosis by anchoring Bcl-2 and Bcl-xL to mitochondrial membranes. Mouse FKBP38 shares 94% and 87% sequence identity with human and rat FKBP38, respectively.\n\nApplications: \nSuitable for use in Western Blot. Other applications no tested.\n\nRecommended Dilution: \nWestern Blot: 0.5ug/ml using HepG2 human hepatocellular carcinoma cell line, NIH-3T3 mouse embryonic fibroblast cell line and L6 rat myoblast cell line\nOptimal dilution to be determined by researcher.\n\nHybridoma: \nProduced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, E. coli-derived, recombinant human FK506 Binding Protein-38 (rhFKBP38; aa 1-325; Accession # Q14318).\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
