Heat Shock Cognate Protein 70, Luminal, Endoplasmic Reticulum (HSC70, BiP)

The endoplasmic reticulum (ER) luminal Hsc70 protein, also known as BiP, is a member of the multigene 70kD family of stress proteins which is involved in the proper translocation and processing of secretory proteins passing through the ER. In spinach, BiP is expressed constitutively in all tissues, except dry seeds, and its levels remain constant in response to environmental stresses such as cold acclimation, drought stress or heat shock (2). This is in contrast to spinach BiP mRNA levels, however, which increase during cold acclimation and decrease in response to drought stress or heat shock (2). Low temperatures may alter protein biogenesis resulting in an accumulation of misfolded proteins that are chaperoned by the existing pool of free BiP protein which in turn, signals an up-regulation of BiP mRNA (2). On the other hand, drought stress and heat shock may reduce protein export through the secretory pathway, thus signaling a down-regulation of BiP mRNA (2). In any case, it appears that the level of constitutively expressed BiP protein is sufficient to maintain proper protein biogenesis when faced with environmental stresses (2).\n \nApplications: \nWestern Blot (Colorimetric) (2,6): 1ug/ml \nWestern Blot (ECL) (7) \nImmunoprecipitation (5): 10ug/ml \nOptimal dilutions to be determined by researcher.

This affinity purified antibody Detects a ~78kD protein, corresponding to the apparent molecular mass of Hsc70 plant BiP on SDS-PAGE immunoblots, in samples from apple, asparagus, carrot, corn, lettuce, orange, potato, spinach, tomato, Arabidopsis thaliana leaves, seeds and siliques, woody plant bark tissue and tobacco. Although this antibody can be used for immunoprecipitations under native conditions, it is not recommended for immunoprecipitating BiP from tomato lysates. This antibody does not cross react with E. coli DnaK, mammalian Hsc70, recombinant hamster BiP or spinach cytosolic Hsc70.
