Hepatocyte Growth Factor, Recombinant, Human (HGF, Scatter Factor, SF, Hepatopoietin A)

Hepatocyte Growth Factor (HGF) is a multifunctional growth factor which regulates both cell growth and cell motility. It exerts a strong mitogenic effect on hepatocytes and primary epithelial cells. HGF synergizes with Interleukin-3 and GM-CSF to stimulate colony formation of hematopoietic progenitor cells in vitro. It may modulate hematopoiesis.\nRecombinant Human HGF is a heterodimer, non-glycosylated, polypeptide chain consisting an alpha-chain of 463aa and beta-chain of 234aa having a total molecular mass of 80kD. \n\nSequence:\nAgrees with the seqeunce of native human HGF.\n\nEndotoxin: \n(same/less than) 0.1ng/ug (IEU/ug) of HGF.\n\nDimers and Aggregates: \n< 1% as determined by silver-stained SDS-PAGE gel analysis.\n\nBiological Activity: \nHepatocyte Growth Factor is fully biologically active when compared to standards. The ED50, calculated by the dose-dependent proliferation of monkey 4MBr-5 indicator cells, was found to be 20-40ng/ml. \n\nProtein Content:\nProtein quantitation was carried out by two independent methods: \n1. UV spectroscopy at 280nm using the absorbency value of 1.83 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). \n2. Analysis by RP-HPLC, using a standard solution of HGF as a Reference Standard.\n\nStorage and Stability:\nLyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile dH2O, 0.1% HSA or BSA. Aliquot and store at -20 degrees C. Reconstituted product is stable for 24 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
