Histone H3, Dimethyl (Lys9)

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.\n\nApplications: \nSuitable for use in ELISA and Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot: 1:500-1:5000 detects dimethyl Histone H3 on 10ug of HeLa acid extract but not on recombinant Histone H3. HeLa acid extract were resolved by electrophoresis, transferred to PVDF membrane and probed with H5110-14E (1:500). Proteins were visualized using a donkey anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nHeLa, NIH/3T3 or COS cell extracts\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
