Histone H3, trimethyl (Lys27)

The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. Originally thought to function as a static scaffold for DNA packaging, histones have now been shown to be dynamic proteins, undergoing multiple types of post-translational modifications, including acetylation, phosphorylation, methylation and ubiquitination (1). \n\nApplications: \nSuitable for use in Immunofluorescence, Western Blot, Immunoprecipitation, Chromatin Immunoprecipitation and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nImmunofluorescence (IC): 1:400\nWestern Blot: 1:1000\nImmunoprecipitation: 1:50\nChromatin Immunoprecipitation: 1:25\nImmunohistochemistry (paraffin): 1:100\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Additional Info

Recognizes endogenous levels of human histone H3 only when tri-methylated on Lys27. Does not crossreact with non-methylated, mono-methylated or di-methylated Lys27. In addition, the antibody does not crossreact with mono-methylated, di-methylated or tri-methylated histone H3 at Lys4, Lys9, Lys36 or Histone H4 at Lys20. Species Crossreactivity: human, mouse, rat and monkey.
