HMG17, HMGN2 (High Mobility Group (HMG) Transcription Factor)

The high mobility group (HMG) chromosomal proteins, which are common to all eukaryotes, bind DNA in a non-sequence-specific fashion to promote chromatin function and gene regulation. They interact directly with nucleosomes and are believed to be modulators of chromatin structure. They are also important in activating a number of regulators of gene expression, including p53, Hox transcription factors and steroid hormone receptors, by increasing their affinity for DNA. Chromosomal HMG proteins have a global fold of three helices stabilized in an `L-shaped' configuration by two hydrophobic cores. Binding: The HMG domain binds to an AT rich DNA sequence using a large surface on the concave face of the protein, to bind the minor groove of the DNA. This bends the DNA helix axis away from the site of contact. The first and second helices contact the DNA, their N-termini fitting into the minor groove whereas helix 3 is primarily exposed to solvent. Partial intercalation of aliphatic and aromatic residues in helix 2 occurs in the minor groove.\n\nApplications: \nSuitable for use in Western Blot and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:500. Detects HMGN2/HMG-17 from acid extracts of murine 10T and HeLa cells, in which a doublet (splice varient) was detected. Acid extracts of HeLa cells were resolved by electrophoresis, transferred to nitrocellulose and probed with H6206. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. \nImmunocytochemistry: This antibody has been reported to show positive immunostaining for HMGN2/HMG-17 in Hep2 and 3T3 cells fixed with 2% paraformaldehyde. \nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
