IL-7 (Interleukin-7, IL7) (Biotin)

IL-7 is a growth factor that largely affects lymphoid development. It appears to act on both immature B and T cells in mice, but in humans it seems to only affect T cells. The receptor complex consists of an IL-7 Ra chain/CD127, which has a MW of 75-79kD, and a common gamma (gc) chain, which is shared by various other cytokine receptors such as IL-2, IL-4, IL-9, and IL-15. IL-7 binding occurs through the IL-7 Ra chain, while signal transduction results following the interaction of the intracellular domains of the IL-7 Ra and gc chains. Soluble forms of IL-7 Ra have been reported. IL-7 Ra is thought to be important in delivering survival signals to thymocytes by up-regulating Bcl-2 expression, as well as promoting the development of intestinal lymphocytes. Cells reported to express IL-7 Ra include double negative thymocytes, CD4+ and CD8+T cells, CD34+ bone marrow cells, epithelial cells, and intraepithelial lymphocytes. Several reports indicate that T cell activation leads to a down-regulation of IL-7 Ra/CD127 expression.1-3.\n\nApplications:\nSuitable for use in ELISA and Western Blot. Other applications not tested.\n\nRecommended Dilution:\nELISA: To detect Human IL-7 by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.25-1ug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with Polyclonal Anti-Human IL-7 as a capture antibody, allows the detection of at least 0.2-0.4ng/well of recombinant Human IL-7.\nELISA (Direct): To detect Human IL-7 by direct ELISA (using 100ul/well antibody solution) a concentration of 0.25-1ug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2-0.4ng/well of recombinant Human IL-7.\nWestern Blot: To detect Human IL-7 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. When used in conjunction with compatible secondary reagents, the detection limit for recombinant hIL-7 is 1.5-3ng/lane, under either reducing or non-reducing conditions.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
