Insulin Regulated Aminopeptidase (IRAP, Placental Leucine Aminopeptidase, PLAP, Leucyl Cystinyl Aminopeptidase, LNPEP, Oxytocinase, Otase, Vesicle Protein of 165kD, vp165)

IRAP (also known as LNPEP) was originally described as an insulin-responsive aminopeptidase found in GLUT4-containing vesicles (1), but later found to be a critical enzyme that regulates the expression and activity of several essential hormones and regulatory proteins, including the GLUT4 transporter (2,3). This membrane associated, zinc-dependent cystinyl aminopeptidase acts as both a receptor for angiotensin IV as well as the enzyme that catalyzes the synthesis of this essential hormone from its angiotensinogen precursor (4). IRAP catalyzes the hydrolysis of several peptide hormones, including oxytocin and vasopressin (3). Abnormal IRAP expression or activity is associated with several forms of cancer in humans, including renal and endometrial cancer (5,6). \n\nApplications:\nSuitable for use in Western Blot, Immunoprecipitation and Immunofluorescence (IF-IC). Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:1000. Incubate membrane with diluted antibody in 5% w/v BSA, 1X TBS, 0.1% \n Tween-20 at 4 degrees C with gentle shaking, overnight. \nImmunoprecipitation: 1:50.\nImmunofluorescence (IF-IC): 1:50 \nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
