Integrin aVB1 is one of five aV and twelve B1 containing Integrin family adhesion receptor heterodimers (1-3). The non-covalent heterodimer of 170kD aV and 130kD B1/CD29 is present on cells that express both subunits, and dimer formation is dependent on the availability of the individual subunits (4). Since the aV and B1 subunits are widely expressed, the aVB1 heterodimer potentially forms in many cell types. The 962 aa human aV extracellular domain (ECD) shares 92-95% aa sequence identity with mouse, rat and bovine aV, while the 708 aa human B1 ECD shares 92-96% aa sequence identity with mouse, rat, bovine and feline B1. The aV ECD contains an N-terminal B-propeller structure, followed by domains termed thigh, calf-1 and calf-2 (1). The B1 ECD contains a vWFA domain, which participates in binding. Each subunit then has a transmembrane sequence and a short cytoplasmic tail. The dimer is folded when it is least active. Divalent cations and intracellular (inside-out) signaling convert it to its most active, extended and open conformation (1). aV integrins bind ligands that contain an RGD motif, including vitronectin, fibronectin and osteopontin (4-9). The relatively weak binding affinity of aVB1 to vitronectin and fibronectin is thought to facilitate its activity in cyclic binding and release during cell migration (4, 5). In oligodendrocytes, astrocytes and pancreatic B cells, aVB1 is expressed early in differentiation when cells are migrating and is down-regulated when differentiation is complete (5-7). aVB1 has also been found to be a receptor for angiopoietin-2 in Tie2-deficient glioma cells, and to mediate cell entry of viruses such as foot-and-mouth disease virus and human metaneumovirus (10-12).\n\nSource:\nRecombinant non-covalent heterdimer corresponding to aa31-992 of human Integrin aV, acidic tail GSL at N-terminal and HHHHHH at C-terminal Accession # (NP_002201) and aa21-728 of human Integrin B1, basic tail GSL at N-terminal Accession # (P05556).\n\nMolecular Weight: \n~115.4kD (aV subunit) 86.4kD (B1 subunit)\n\nEndotoxin Level:\n(same/less than)1EU/1ug (LAL)\n\nBilogical Activity:\nMeasured by its binding ability in a functional ELISA. When recombinant human Integrin aVB1 is coated at 5ug/ml, Human Fibronectin binds with an apparent KD < 0.1nM.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
