Interleukin 13 (IL-13)

IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and also by mast cells and NK cells. Targeted deletion of IL-13 in mice resulted in impaired Th2 cell development and indicated an important role for IL-13 in the expulsion of gastrointestinal parasites. IL-13 exerts anti-inflammatory effects on monocytes and macrophages and it inhibits the expression of inflammatory cytokines such as IL-1b, TNF-a, IL-6 and IL-8. IL-13 has also been shown to enhance B cell proliferation and to induce isotype switching resulting in increased production of IgE. Blocking of IL-13 activity inhibits the pathophysiology of asthma. Human and murine IL-13 is cross-species reactive. Recombinant human IL-13 is a 12.5kD protein consisting of 114 amino acid residues.\n\nApplications:\nSuitable for use in Direct ELISA, Western Blot and Neutralization. Other applications not tested.\n\nRecommended Dilutions:\nNeutralization: 0.12-0.15ug/ml to yield one-half maximal inhibition [ND50] of the biological activity of hIL-13 (2ng/ml).\nDirect ELISA: 0.5ug/m using 100ul/well antibody solution. The detection limit for rhIL-13 is 0.2-0.4ng/well.\nWestern Blot: 0.1-0.2ug/ml. The detection limit for rhIL-13 is 1.5-3.0ng/lane, under either reducing or non-reducing conditions.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
