IP-10 (Cxcl10, C-X-C Motif Chemokine 10, 10kD Interferon gamma-induced Protein, Gamma-IP10, Interferon-inducible Protein 10, Protein Mob-1, Small-inducible Cytokine B10, Inp10, Mob1, Scyb10)

IP-10 is a highly inducible, primary response gene that belongs to the CXC chemokine superfamily. It was first cloned in 1985. Like Mig, IP-10 has no activity on neutrophils. Its functions include stimulation of monocytes, natural killer and T-cell migration, regulation of Tcell and bone marrow progenitor maturation, modulation of adhesion molecule expression as well as inhibition of angiogenesis. Mig and IP-10 share the same receptor, CXCR3.\n\nApplications:\nSuitable for use in ELISA, Western Blot, Immunohistochemistry and Neutralization. Other applications not tested.\n\nRecommended Dilution:\nELISA: To detect Rat IP-10 by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.5-2ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Rat IP-10 as a detection antibody, allows the detection of at least 0.2-0.4ng/well of recombinant Rat IP-10.\nWestern Blot: To detect Rat IP-10 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Rat IP-10 is 1.5-3ng/lane, under either reducing or non-reducing conditions.\nImmunohistochemistry: This antibody stained colchicine injected rat brain (including the caudate putamen) tissue. The primary antibody was incubated at 0.25ug/ml overnight at 4?C. This was followed by a peroxidase conjugated secondary antibody and then a fluorescein Tyramide Signal Amplification reagent.\nNeutralization: To yield one-half maximal inhibition of the biological activity of Rat IP-10 (100ng/ml), a concentration of 5-10ug/ml of this antibody is required.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
